摘要:

以国产正置显微镜为平台搭建简易可行的光镊装置,实现了对单个微球的二维捕获,并将其与荧光检测技术相结合,以性质优良的量子点(荧光发射波长605nm)为荧光标记物,建立了一种基于单个微球富集待测分子的高灵敏度高选择性的定量分析方法,实现了人乳头瘤病毒16的DNA的定量分析.

Abstract:

A simple and feasible optical tweezers setup based on a domestic upright fluorescence microscope was constructed, and it was successfully applied for two-dimensional trapping of a single microsphere. By combining this setup with fluorescence detection and using fluorescent quantum dots (QDs) with emission wavelength 605nm as the tags, a quantitative method with high sensitivity and selectivity was developed when single microspheres were applied to enrich the target molecules, and as a proof-of-concept study the quantitative analysis of human papillomavirus (HPV) 16 DNA was realized.
